Fibrogenic and fibrolytic potential of differently activated human macrophages

چکیده
Macrophages are involved in the regulation of fibrogenesis and turnover extracellular matrix. One way to perform this function is through production profibrotic fibrolytic factors including fibronectin, laminin, collagen, matrix proteases. The most them has been well studied experimental models; however, much remains unclear regarding human macrophages. Therefore, aim study was content proteases (MMP-2 MMP-9, cathepsin L), their inhibitors (TIMP-1), collagen (type I) supernatants differently activated We compared macrophages differentiated by M-CSF or GM-CSF further polarized M1 with lipopolysaccharide, M2a IL-4, M2c dexamethasone. obtained from peripheral blood monocytes. MMPs, TIMP, cathepsin, determined using appropriate ELISA kits. results demonstrate that differentiation more important for above polarizing stimuli (lipopolysaccharide, dexamethasone). Moreover, showed predominantly antifibrotic activity because pronounced MMPs production, while GM-CSF-induced cultures, on contrary, were characterized properties due high level TIMP-1 type I collagen. M1, M2a, M2c, induced M-CSF, differed only MMP-2 produced metalloproteinase than other subtypes. In case GM-CSF-differentiated cells, a higher and, lesser extent, characteristic whereas have minimal concentration among macrophage Concerning L relatively constant did not depend generation conditions (differentiation signals). Thus, data help identify subtypes anti- potential may be useful development cell therapy diseases associated dysregulation.

Human activated macrophages and hypoxia: a comprehensive review of the literature
Macrophages accumulate in poorly vascularised and hypoxic sites including solid tumours, wounds and sites of infection and inflammation where they can be exposed to low levels of oxygen for long periods. Up to date, different studies have shown that a number of transcription factors are activated by hypoxia which in turn activate a broad array of mitogenic, pro-invasive, pro-angiogenic, and pro...

Human macrophages derived from in vitro culture of peripheral blood monocytes were studied under a variety of conditions to determine their microbicidal capacity for the obligate intracellular protozoan, Toxoplasma gondii. The effect of macrophages on intracellular Toxoplasma was evaluated morphologically by light and phase microscopy and by autoradiography. When macrophages from dye test (DT)-...

The digestion of dietary fibers is a major function of the human intestinal microbiota. So far this function has been attributed to the microorganisms inhabiting the colon, and many studies have focused on this distal part of the gastrointestinal tract using easily accessible fecal material. However, microbial fermentations, supported by the presence of short-chain fatty acids, are suspected to...

In order to determine if human LAK cells were cytotoxic against autologous AM phi, we studied the ability of human peripheral blood MNCs, stimulated in vitro with recombinant human IL-2, to lyse AM phi in a four-hour 51Cr-release assay. These cells showed significant cytotoxicity against autologous AM phi. The AM phi which had been cultured for four days served as better targets than freshly is...
